C02, Groß, will continue work on cytokine-mediated impairment of macrophage homeostasis resulting in inflammatory consequences. The focus will be on the consequences of a lack of IFNγ and of GM-CSF-induced signaling on macrophage activity. In addition to effects on the NLRP3 inflammasome, the focus will be broadened to elucidate the impact on macrophage metabolism and the epigenetic landscape in general, using unbiased approaches. The role of these mechanisms in SARS-CoV-2 immunopathology triggered by a lack of IFNγ and a proposed pathologic feed-forward mechanism between T cell-derived GM-CSF and NLRP3-dependent IL-1β from macrophages will be studied in close collaboration with C01.
