Chronic inflammatory diseases form a highly complex, interdisciplinary field of research. A key technology for the investigation of inflammatory processes is the phenotyping and spatial representation of specific cell types in their respective disease- and tissue-specific microenvironment. Mass cytometric tissue analysis combines for the first time the advantages of mass cytometry with the possibility of spatial representation in tissues. Mass cytometry uses antibodies coupled with rare elements for antigen detection. The minimal signal overlap of mass cytometry allows for simultaneous analysis of up to 50 parameters in one slide. cyclometric tissue analysis enables a highly detailed analysis of multiple parameters directly in context with the local microenvironment. A comparable technology is currently not available at the University Hospital Erlangen. The equipment would enable the applicants to perform detailed and multidimensional characterizations of different cell populations and of their interaction directly in inflammatory tissues. It will further expand the possibilities of the established Core Facility for Imaging and enrich the priority focus of inflammation research established in Erlangen.
