Project P20 investigates how sphingolipids (SL) are transported in the endolysosomal system and how sphingolipid homeostasis is maintained. Although SLs are enriched at the plasma membrane they also play important roles in other organelles, such as the Golgi apparatus or vacuoles in yeast. The Golgilocalized GARP complex has an essential role in this process. To address how sphingolipids are sorted into and out of the yeast vacuole, the group will (i) determine the vacuolar sphingolipid content, (ii) identify vacuolar sphingolipid binding/transport proteins, and (iii) uncover the molecular mechanisms of vacuolar sphingolipid binding/transport proteins.
