The NLRP3 inflammasome is a major instigator of disease-associated inflammation and immunepathology in many tissues including the CNS. However, little is known about the role of NLRP3 inmaintaining homeostasis. Combining murine ex vivo CNS culture systems, in vivo models of resolvingneurological injury, and inducible genetic models of activating NLRP3 patient mutations, we willcharacterize NLRP3 activation and its consequences in complex CNS tissue and study the earlyevents of pathogenesis of NLRP3-driven neuroinflammation that might guide therapeutic interventionsin diseases such as Multiple Sclerosis and Alzheimer's disease or Parkinson's disease.
