Sustaining synaptic transmission over long periods of time is crucial for the function of the nervous system. The proteins mediating presynaptic neurotransmitter release are being intensely investi-gated. However, the energy homeostasis of the presynapse is less well understood. To address this fundamental question, we will combine high-resolution time-lapse imaging of ATP levels and other metabolites with electrophysiological techniques at cerebellar mossy fiber boutons in acute brain slices. The project has the potential to revealed the mechanisms controlling energy homeo-stasis of the presynapse and potential energetic constraints of transmitter release.
