The main aim is to discover and functionally dissect the molecular mechanisms during differentiation and disease development of:A) DAXX-dependent H3.3-mediated silencing of heterochromatic genes and the chaperone dependence of H3.3K27M in tumor development, and B) the PWWP2A-interactome with a functional focus on the novel “core NuRD” and the proposed PHF14-RAI1-TCF20-HMG20A (PRTH) complexes in transcriptional regulation of PWWP2A-bound H2A.Z target genes.
