We will study, both experimentally and theoretically, hybridization kinetics within oligonucleotide pools and investigate how the composition of the pool – single-strands, partially hybridized, non-cognate complexes, and “perfectly matched” complexes – changes over time. As hybridization is strongly dependent on environmental parameters such as salt concentration and pH, we will then study the spatiotemporal dynamics of the sequence pools within salt gradients. We will develop experimental methods to monitor different species within the pool at different locations, and theoretical tools to analyze these complex dynamics.
