Unraveling the differential role of myeloid cells in the regulation of ANCA-induced glomerulonephritis

The aim of the research proposal is the detailed analysis of the pathogenic contribution of different myeloid subspecies and the identification of a regulatory network between myeloid cells and T-cells in ANCA-induced glomerulonephritis. We and others have shown that both neutrophils and monocytes are activated in vitro by ANCA. However, their contribution to the induction of an ANCA-glomerulonephritis and the specific role of different myeloid subpopulations is not understood. Therefore, we will analyze the contribution of different myeloid subpopulations to ANCA-glomerulonephritis by specific modulation of these subpopulations in two different ANCA-animal models. Furthermore, we will characterize a regulatory network between myeloid cells and adaptive T-cell immunity in ANCA-glomerulonephritis. In addition, we aim to establish a urinary myeloid and T-cell flow-cytometry signature in patients with ANCA-glomerulonephritis. In summary our studies will result in a better understanding of the role of myeloid cells in the pathogenesis of ANCA-glomerulonephritis and their role in the regulation of T-cell immunity.
