We will evaluate the therapeutic performance of combined modality radiation therapy with radiosensitizers and multi-functional antibody derivatives aimed at modulating innate and adaptive immune checkpoints. Radiosensitizing candidate drugs will be identified in screening approaches followed by mechanistic characterization. Basal and therapy-induced vulnerabilities in immune checkpoints will be examined and exploited using multi-functional antibody derivatives. Combinations of the most promising approaches will be analyzed in syngeneic transplantation models and investigated at the cellular, transcriptomics, and proteomics level.
