Sequence-specific manipulation of mRNA provides access to studying and controlling gene expression at the post-transcriptional level which is of particular importance for molecular biology and cellular applications. The 5′ cap is present in all eukaryotic mRNAs and m6A is the most abundant internal modification in mRNA. Both modifications control numerous interactions but currently cannot be installed or removed in a sequence-specific manner. This project aims to obtain sequence-specific mRNA manipulation at these modifications by creating fusion constructs of RNA-modifying enzymes with RNA-binding proteins or RNA-protein complexes. The precise manipulation of modifications will be used to label or isolate a specific transcript and to evaluate biological effects, particularly translation efficiency and stability.
