We identified a population of specialized regulatory antigen presenting cells, termed tissue-resident macrophages, in the gastro-intestinal tract that protects from acute GvHD. These intestinal macrophages control pathogenic T cells via PD-L1. By employing preclinical mouse models for GvHD and GvL and analyses of allo-HSCT patient samples, we will interrogate the molecular cues that drive immunoprotective antigen presenting cell differentiation and function. We will explore these cells in tissues of GvHD such as the intestinal tract and skin and compare them with tissues, where GvHD typically does not manifest. Finally, potential prophylactic and therapeutic strategies will be tested.
