Electron microscopy (EM) is the only technique which allows to image the arch¬itecture of the TM system, there is simply no other way to visualize the complexity of this membrane system and also localize proteins therein at the nanoscale. Questions regarding TM ultrastructure in mutants or different developmental stages and the localization of proteins are central for all groups of the Research Unit. We will provide skilled technical support for transmission electron microscopy for all groups to help them answer their questions regarding TM ultrastructure and spatial organization of proteins therein. During the first funding period we did establish a high through-put workflow for the analysis of TM topology using chemically fixed and plastic embedded samples. Such ultrastructural analyses using optimized protocols will play an important role also during the second funding period and will be extended by a workflow for the localization of proteins by immunogold techniques.
