Processing of the amyloid precursor protein by the ß-secretase and the γ-secretase module leads to the generation of amyloid ß-peptides and a soluble intracellular C-terminal domain, which is involved in transcriptional regulation. The goal of this project is stepwise to reconstitute the enzyme/substrate complex and to structurally analyze the interaction of multiple transmembrane segments by pulsed electron–electron double resonance spectroscopy based on a GxxxG motif which was found to mediate helix-helix interactions within subunits of the γ-secretase module.
