Understanding of modulation and regulation of proteasomal activity cannot be complete without structural information about proteins involved. So far structure, function and mechanism have been elucidated only for 20S proteasome and few regulatory components. Our aim is to determine the structure of the lid and the signalosome complex, playing major role in the first steps of recognition and degradation of folded proteins. We want to combine and expand structural methods for characterization of major compounds of the protein degradation machinery and to provide mechanistic and modular insights into its regulation.
