Mass spectrometry imaging (MSI) uses spatially resolved proteomic and metabolomic techniques to illustrate the simultaneous spatial distributions of hundreds of biomolecules directly on patient tissue samples. Last years the importance of the MALDI (Matrix-assisted laser desorption/ionization)-Imaging in tissue analysis especially in brain tissue exploration has increased significantly, and its advantage for the exploration of the brain proteome changes that accompany the neurodegenerative disease have been shown in different studies. The samples can be directly analyzed; no need of additional extraction steps of compounds and the labeling of the sample is dispensable. The spatial resolution of mass spectrometry instruments is one of the limiting factors in advancing the mass spectrometric tissue imaging. The laser beam diameter enables high spatial resolution, moreover, the soft ionization of the laser allows the detection of intact proteins and qualifies this instrument for molecular imaging and for clinical research ambitions.
