The assembly of the C-ring, a crucial cytoplasmic part of the bacterial flagellum, can be reconsti-tuted in vitro in the presence of FlhF and its regulator ATPase FlhG for the S. putrefaciens system. We will continue to elaborate this in vitro system to dissect the C-ring assembly process and characterize the involvement of FlhF and FlhG by cryo-electron microscopy (cryo-EM). In addition, we will purify and characterize mainly by cryo-EM one of the crucial polar landmark proteins, HubP, which establishes correct spatiotemporal patterns in the monotrichous S. putrefaciens, for example by recruiting FlhF to the pole region.
