This project aims at elucidating the molecular principles underlying spatio¬temporal coordination of genomic DNA replication and segregation with cell growth and division in Sinorhizobium meliloti. Specifically, we will study (i) assembly of Rgs (rhizobial growth and septation) proteins at cell division site and newest cell pole, (ii) the role of the polar scaffold proteins RgsE and PopZ in integrating cell elongation and chromosome segregation, (iii) factors involved in spatially confining the oris of the three replicons in the polar and subpolar areas, (iv) the mechanism of RepABS partition complex formation, and (v) the role of GcrA and CcrM-mediated methylation in regulating replication initiation of the secondary replicons.
