We will (i) analyze functional changes of microglia harboring alterations in TREM2, known to be a risk factor Alzheimer's disease (AD), in ex vivo assays, (ii) assess whether AD pathology in A-overexpressing mice harboring myeloid cell-/microglia-specifically TREM2 alterations is changed and (iii) identify the genomic signature of such microglia. We will also dissect the molecular and operating determinants and molecules of microglia effector functions in A, immunotherapy, which - upon identification - will be validated by in vitro assay, and, ultimately in vivo with respect for their impact on modulating AD pathology.
