Project A03 focuses at obtaining a deeper understanding of the processes involved in presynaptic active zone (AZ) remodeling, which control the release of synaptic vesicles. To this end we will employ simulated emission depletion (STED) and single molecule super-resolution fluorescence microscopy, as well as electron microscopy (EM) and cryogenic electron tomography (cryo-ET) to analyze conserved cytomatrix proteins architectures. Ultimately, we aim to arrive at a description of the AZs macromolecular scaffold organization and its dynamics during adaption in the (sub-)nanometer range.
