We intend to uncover the high light and cold responsive small non-coding RNA (sRNA) repertoire in A. thaliana and to study their role in the regulation of genes encoding plastid-localised proteins. We will quantify levels of sRNAs and their cognate targets by RNA-seq in response to the high light and cold treatments that are centrally studied in the CRC/Transregio. We will also quantify sRNA and target RNA levels by RNA-seq in selected A. thaliana mutants with disordered retrograde signalling. By imposing perturbations to the identified plastid-related sRNA-regulated circuits we will examine their role in plant acclimation to high light and cold.
