This project targets proteins that link endosomal pathways and autophagy networks. One prime candidate is PLEKHM1 whose role in regulation of autophagosome-lysosome fusion will be analyzed. In addition, we will systematically screen for Rab GAP proteins that coordinate the core autophagy and membrane trafficking machineries. Functional screening will be performed using the CRISPR/Cas9 technology. Moreover, we will dissect the functional role of the Rab GAP TBC1D5 in autophagy, clathrin-mediated endocytosis, and neu-roendocrine signaling using C. elegans as model organism.
