The preliminary data of project A03 indicates that glucocorticoid-mediated anti-inflammatory and pro-resolving effects rely on a reprogramming of mitochondrial metabolism and the production of the anti-inflammatory metabolite itaconate. We seek to better dissect the metabolic nexus of glucocorticoid receptor signaling and mitochondrial activity in macrophages. We will study the relevance of metabolic reprogramming for the anti-inflammatory and pro-resolving effects of glucocorticoids in vitro as well as in preclinical models of immune-mediated diseases and in patients.
