RAGE mediated DNA repair at the interface between damage by reactive metabolites and repair of diabetic complications (C06+)

In diabetes type 1 and type 2 DNA double-strand breaks accumulate due to a reduced ability to repair DNA, resulting in senescence and ultimately fibrosis of an organ. RAGE is translocated into the nucleus, phosphorylated by ATM. RAGE forms the MRN complex and alters the structure of the DNA, so that MRE-11 can become active. Reconstitution of RAGE into a diabetic mouse leads to a remission of existing late complications. The project investigates how RAGE inhibits diabetes associated DNA damages and reverses late complications.
