GRK 2098: Biomedizin des saure Sphingomyelinase/saure Ceramidase Systems
Zusammenfassung der Projektergebnisse
Das Graduiertenkolleg 2098 (GRK-2098) untersuchte verschiedene Aspekte der Biomedizin von Sphingolipiden. Sphingolipide spielen eine zentrale Rolle bei vielen zellulären Prozessen, wie z.B. Proliferation, Zelldifferenzierung und Zelltod, aber u.a. auch bei bakteriellen und viralen Infektionen, zystischer Fibrose, Pneumonien, Multipler Sklerose, kardiovaskulären Erkrankungen, Krebs, majorer Depression, neurodegenerativen Erkrankungen bzw. Makuladegeneration. Das GRK 2098 konzentrierte sich auf Untersuchungen zur Rolle von Sphingolipiden bei Infektionserkrankungen, im Immunsystem, bei Auto-Immunerkrankungen, Krebs und kardiovaskulären Erkrankungen. Alle Projekte untersuchten die Rolle des saure Sphingomyelinase/Ceramid/saure Ceramidase/Sphingosin/Sphingosin-kinase/Sphingosin 1-phosphat (S1P) Systems bei den genannten Erkrankungen. Neben den wissenschaftlichen Arbeiten organisierte das GRK 2098 ein umfangreiches Lehr- und Ausbildungsprogramm im Bereich "Biomedizin von Sphingolipiden" für die beteiligten PhD-, MD/PhD-, Dr. med.-Doktoranden/innen und Abiturienten/innen. Arbeiten des GRK 2098 zeigten eine völlig neue Funktion von Sphingosin als anti-viraler Abwehrmechanismus in multivesikulären Körperchen. Sphingosin bindet an Viren, verhindert die Freisetzung ins Zytoplasma und vermittelt lysosomalen Abbau des Virus. Diese Studien beschreiben einen völlig neuen Mechanismus zur infektionsbiologischen Rolle von Sphingolipiden und wurden in Nature Communications publiziert. Weitere wichtige Arbeiten des GRK 2098 konnten zeigen, dass die Expression der sauren Sphingomyelinase eine wichtige Rolle bei der Abwehr intestinaler Infektionen mit Citrobacter rodentium spielt. Defizienz der sauren Sphingomyelinase führte zu einer überschießenden Infektion, einer unkontrollierten Th1 and Th17 Antwort und einer schweren Colitis. Diese Studien wurden in Front. Immunol. publiziert. Versuche mit dem Parasiten Plasmodium yoelii zeigten ebenfalls eine protektive Rolle der sauren Sphingomyelinase bei Infektionen: Eine erhöhte (transgene) Expression der sauren Sphingomelinase spezifisch in T Lymphozyten schützte in einem Mausmodell vor der Entwicklung einer Parasitämie. Auch diese Studien wurden in Front. Immunol. publiziert. Weitere Untersuchungen zur Rolle der sauren Sphingomyelinase im Immunsystem ergaben, dass das Enzym eine wichtige Funktion bei der Regulation der Aktivierung und Proliferation von CD4+ T Zellen, insbesondere von regulatorischen T Zellen in vivo, hat. Die erhöhte Zahl immunosuppressiver regulatorischer T Zellen in saure Sphingomyelinase-defizienten Mäusen könnte auch Befunde des GRK 2098 erklären, dass saure Sphingomyelinase-defiziente Mäuse vor der Entwicklung von Auto-Immunkrankheiten, insbesondere einer Auto-Immunarthritis, geschützt sind. Andererseits führt der Abbau von Ceramid zu Sphingosin 1-phosphat zu einer Migration pro-inflammatorischer Immunzellen z.B. in die Orbita bei Graves' Orbitopathie. Diese Studien wurden in Thyroid and Invest. Ophthalmol. Vis. Sci. publiziert. Die verschiedenen Studien des GRK zeigen, dass Sphingolipide sowohl anti- als auch proinflammatorische Funktionen haben und dass das Gleichgewicht von Ceramid, Sphingosin und Sphingosin 1-phosphat eine wichtige Rolle bei der Regulation des Immunssystems und von antibakteriellen, -viralen und -parasitären Immunantworten spielt. Projekte des GRK 2098 zu kardiovaskulären Funktionen von Sphingolipiden konnten eine völlig neue Funktion von Sphingolipiden bei Vaskuloneogenese nach einem Schlaganfall zeigen. Diese Untersuchungen wiesen nach, dass funktionelle Inhibitoren der sauren Sphingomyelinase die Vaskuloneogenese nach einem ischemischen Schlaganfall fördern. Weitere Studien des GRK 2098 konnten eine wichtige Rolle der sauren Sphingomyelinase bei Tumormetastasierung, der Aktivierung von Makrophagen durch Mykobakterien und der Wirkung von Toxinen nachweisen. Grundlagenwissenschaftliche Projekte beschäftigten sich mir der Rolle von Sphingosin 1-phosphat und den entsprechenden Rezeptoren beim Transport von Cholesterol und der Pathogenese von Aortenaneurysmen. Das Ausbildungsprogramm des GRK 2098 vermittelte eine intensive Ausbildung in der Biologie und der Biomedizin von Sphingolipiden. Das GRK 2098 bot eine große Zahl verschiedener Lehrangebot an, wie z.B. regelmäßige Vorträge und Präsentationen, Forschungstage, intensive Diskussionen zu den Promotionen, Teilnahme an wissenschaftlichen Kongressen und die Möglichkeit, bis zu 12 Monaten an einem Labor unserer Partner in New York wissenschaftlich zu arbeiten.
