The project aims at dissecting aspects of HIV spread and pathogenesis that are not accessible to experi-mental investigation in classical cell culture models. We previously established that 3D extra-cellular envi-ronments shape the mode and efficacy of HIV spread in lymphocyte cultures and that HIV-1 suppresses the helper function of infected CD4 T cells to B cells. We will now (i) integrate dendritic cells to assess the im-pact of 3D environments on the complex physical and functional interactions between HIV-1 and its target cells and (ii) study effects of HIV-1 on CD4 T cell help to CD8 T cells. These analyses will yield new insight in patho-physiological principles of HIV-1 infection.
