Our analysis of the dynamics of early HIV-1 replication focuses on the viral capsid, a central regulator of this stage. We have developed, validated and applied genetic code expansion and click-labelling for direct detection of the capsid protein, and collaborated with Kräusslich on labelling the reverse transcribed ge-nome. We will now pursue live cell imaging, combining capsid and DNA-labelling strategies with labelled host proteins for a detailed characterization of events at and beyond the nuclear pore. Furthermore, we will perform comparative analysis of post-entry dynamics of the retrovirus murine leukemia virus and HIV-1 to elucidate the roles of the morphologically distinct capsids in the context of the different replication strate-gies of these viruses.
