The AAA-ATPase Cdc48/p97 is essential for cellular protein homeostasis and genome stability, thus implicating it in degenerative diseases and cancer; but how it mediates the proposed structural remodelling of critical substrate proteins that underlie its cellular roles is still unclear. We wish to apply supramolecular chemistry combined with newly developed biochemical approaches to clarify how p97 works on the molecular level, hopefully to also reveal new possible strategies for pharmacological intervention.
