In this project we aim at developing a general host-guest complexation strategy for the manipulation of the stability and activity of desired proteins specifically encapsulated into customized DNA cages. To this end, we will use a large tool of supramolecular ligands with distinct specificity, affinity and associated read-out signals. Spatial control of multivalent ligand-protein interactions will be also employed to mimic the architecture of fundamental components of the kinetochore-microtubule interface, thus enabling to gain a deeper insight into the mechanisms of chromosome segregation and more in general to offer a strategy for the generation of artificial protein machineries.
