In project A1 we develop multi-armed peptide based ligands for proteins in order to modulate their function (e.g. by interfering with essential protein protein interactions). The ligands contain non-natural amino acids with tailor-made specific supramolecular binding sites, such as our guanidiniocarbonyl pyrrole cation as a highly efficient anion binder. The ligands therefore target anionic hot spots on the proteins. Ligand design is based on available structural information of the target and for optimization combinatorial approaches such as the screening of dynamic combinatorial libraries is used.
