Nanocapsules, which combine both high drug loading (>80%) and extended release properties, will be prepared by novel physical methods as an alternative to matrix-type nanoparticles. More flexibility in drug release will be achieved by combining different nanocarriers and by pH- and sebum-triggered nanocarriers, which will release the drug at a desired pH-threshold or by dissolution/plasticization in sebum in diseased skin or within hair follicles. In addition, the performance of nanocarriers after incorporation in final dermal vehicles as well as their direct preparation therein and important nanocarrier stability issues after preparation, during storage and after skin administration will be investigated.
