Post-translational modification of cellular proteins with ubiquitin-like SUMO modifiers is a dynamic process, in which SUMO-specific isopeptidass of the SENP family acts as SUMO deconjugases. We could demonstrate that in hypoxia the catalytic activity of distinct SENPs is reversibly inhibited. By comparative proteomics we defined a distinct subset of hypoxia-induced SUMO1 targets, including a set of transcriptional regulators. Based on ongoing functional experiments we propose a pathway that connects oxygen-controlled SENP activity to hypoxic reprogramming of cellular metabolism.
