Ebola virus RNA synthesis is regulated by the interplay of noncoding RNA elements in the genome and viral nucleocapsid proteins, however, mechanistic details are still enigmatic. In the proposed project that intimately combines virological with biochemical expertise, we will investigate (i) the role of RNA elements in the 3'-leader promoter and at internal transcription start sites, (ii) the role of leader transcripts and VP35 helicase activity in viral RNA synthesis, (iii) establish an Ebola virus in vitro transcription system to study viral RNA synthesis and address (iv) how the encapsidated genome can be accessed by the viral polymerase complex.
