Using a mouse model which allows reversible antigen presentation in vivo the features and fates of CD8+ T cells exhaustion solely by antigen will be explored. The aim of the project is to investigate the degree and reversibility of the exhausted state by tissue grafts, autoimmunity induction and viral challenges and by analysing the cells' transcriptome and epigenetic modifications. In addition it will be studied how a modified mouse herpesvirus expressing the same antigen persists without exhausting the respective T cells.
