Monothiol glutaredoxins are central components of cellular iron homeostasis. They are involved in iron uptake regulation, iron trafficking, biogenesis of iron-sulfur proteins, and assembly of proteins with iron centers. How monothiol glutaredoxins fulfil these diverse physiological functions is unclear. To elucidate their molecular function, we will employ cell biological, proteomic, biochemical, and biophysical approaches with the aim to identify new interaction partners and to define the molecular mechanism of these glutaredoxins and of their bound [2Fe-2S] cluster.
