Project A4 addresses the functional role of CDPKs in cold priming of Arabidopsis thaliana. In line with our working hypothesis, cold primed plants display improved tolerance to later cold stress. We have identified one distinct CPK isoform as a negative regulator of cold priming, and cpk mutant plants display enhanced frost tolerance, accompanied by corresponding alterations in transcript-, metabolite-, and lipid-accumulation patterns. Future analysis will investigate the biochemical regulation of the enzyme, and ¿ by assessment of co-expressed genes in a priming/memory context ¿ the identification of transcriptional regulators as potential CPK phosphorylation targets.
