S01 offers a variety of standardized in vivo and in vitro disease models including mouse models and mouse phenotyping as well as human iPSC models – particularly CRISPR-engineered iPSC lines and engineered heart muscle (EHM). With a substantial need for specific reporter models, S01 will expand its portfolio of iPSC models according to the specific demands of the CRC 1002. In addition, methods and protocols will be continuously refined, and novel technologies such as scalable cardiomyocyte and fibroblast production in bioreactors, multi-well plate, highly parallelized EHM disease modelling, as well as base editing and prime editing in iPSC models will be implemented. In S01, we have also established single cell and single nucleus sequencing in myocardial biopsy samples and will support CRC projects applying these techniques.
