A09 develops mass spectrometry techniques to analyze protein interactions in cardiomyocytes and to map the human proteome in cardiac biopsies. We hypothesize that therapeutically relevant Caveolin3 and Junctophilin2 interactors exist, which will be analyzed in the context of subcellular Ca2+ release via Ryanodine Receptor clusters. High-resolution imaging workflows will quantify protein clustering versus fragmentation in human iPSC-cardiomyoctes and in cardiac biopsies. We aim to establish the proteomic and cellular basis of heart failure progression versus functional recovery based on in-depth analyses of excitation-contraction coupling models.
