Cardiomyocytes feature an elaborate membrane system. Little is known about membrane protein biogenesis and its relevance to disease in this system. Schwappach et al. recently co-discovered a pathway involved in the posttranslational membrane integration of C-terminally membrane-anchored (tailanchored, TA) membrane proteins, the TRC40 pathway. We will therefore tackle the consequences of an impaired cardiac TRC40 pathway, e.g. lack of cardiomyocyte TA protein function, as opposed to a dominant negative activity of mistargeted TA proteins, and address the contribution of the TRC40 pathway to the cardiomyocyte proteostasis network.
