Most projects within the research unit will require poly- and mononoclonal antibodies as essential tools to follow up specific project-re levant questions. Polyclonal antibodies can usually be generated by companies (such as Eurogentec or BioGENES) with acceptable quality, whereas the quality of many monoclonal antibodies, especially of those which are commercially available, is often non-satisfactory. Moreover, custom generation of monoclonal antibodies is by far too expensive to be funded within the individual projects. Therefore a centralised service project will be established for the generation of monoclonal antibodies. Other bottle necks in terms of detailed biochemical and structural analysis of isolated proteins and protein complexes concern rapid identification and characterisation of proteins by mass spectrometry and recombinant expression and production of proteins in insect cells using baculovirus. Since the project members have usually not established these techniques within their own laboratory and/or have no direct access to the quite expensive and often complex infrastructure, the research unit intends to establish a centralised service project, to provide the required technologies, i.e. monoclonal antibody generation, mass spectrometry and protein production in insect cells.
