In this project, we will apply a set of single-molecule calibrated tension sensor modules, which were developed and calibrated in the previous funding periods, to generate Förster resonance energy transfer (FRET)-based probes that report piconewton-scale forces across central components of integrin- and cadherin based adhesion structures. By combining these tension sensors with a recently established multiplexing approach, we will simultaneously investigate the molecular mechanics of cell-matrix and cell-cell adhesion during collective cell migration.
