Poly(ADP-ribosyl)ation is the enzymatic posttranslational modification of proteins with poly(ADP-ribose) (PAR), a linear or multibranched polyanion of variable size that can also bind non-covalently to numerous proteins via specific PAR binding motifs. DEK is a highly conserved chromatin architectural factor and a target of both covalent and non-covalent modification by PAR. In this project we propose to investigate how PAR affects human DEK as a model protein. We will focus on functions of DEK which are related to the cellular response to replicative and genotoxic stress. By characterizing PAR-dependent molecular interactions and localizations of DEK at the replication fork and at sites of DNA damage, this project will advance our understanding of the fundamental mechanisms by which PAR regulates proteostasis.
