In project A07 we have shown that the activation and function of intersectin-1 in synaptic vesicle recycling and of SNX9 in endocytosis is controlled by release from intramolecular autoinhibition. In the coming funding period, we will (i) unravel the structural basis of the interaction between intersectin-1 and the SNARE-complex, (ii) investigate the role of intersectin-1 in endocytic protein nanoscale localization and dynamics, and (iii) unravel the formation and function of the early endocytic intersectin-FCHo-Eps15 complex in clathrin-mediated endocytosis.
