We have shown that synaptic vesicles (SVs) are recycled via clathrin-independent actin-mediated endocytosis. Using cell biological and (opto-)genetic approaches together with super-resolution microscopy and modeling we will now dissect how actin-nucleation by formins regulates actomyosin activity to facilitate SV membrane retrieval, how these activities are coordinated with the acquisition of membrane curvature via BAR-domain proteins, and how SVs are reformed from endosomal-like vacuoles by the endocytic scaffolds intersectin 1 and 2.
