Project P10 (Steinhoff) dissects the mechanism of signal transduction in bacterial chemo- and phototaxis via the photoreceptor/transducer NpSRII/NpHtrII of the halophilic archaeon Natronomonas pharaonis, which are clustered in microcompartments of the plasma mem-brane. Taking advantage of EPR spectroscopy, the group will (a) dissect the mechanism of adaptation and dynamic allostery within the signaling complex by comparing the methylated and demethylated states, and (b) analyze the mechanism of Ca2+-ions pumping by channelrhodopsin to understand light induced conformational changes in a related system.
