Project P5 focuses on the organization and coupling/crosstalk of the two major membrane microcompartments during synaptic vesicle (SV) cycling in neurons, the sites of docking and fusion (exocytosis) of the so-called readily releasable pool (RRP) of SVs at the ‘active zone’, and the sites for SV retrieval (endocytosis) at the periactive zone. The group succeeded in formation of functional, purely presynaptic boutons onto glass coverslips, microstructured and functionalized with synaptic cell-adhesion molecules. They will use this new preparation to study aspects of the structure and function of exo- and endocytic microcompartments by super-resolution fluorescence microscopy and correlative SEM.
