The project studies proteolysis in the regulation of non-apoptotic cell death. The mitochondrial serine protease HtrA2/Omi is a component of TNFα-induced necroptosis. Furthermore, an unexpected contribution of ADAM17 in the induction of necroptosis was discovered, which will be further analyzed in the next funding period. The role of caspase-8 in necroptosis is different in human and murine cells since caspase-10, a homolog of caspase-8, is present in humans but not in mice, pointing to a species-specific role of caspase-8 in the regulation of necroptosis.
