The overall objective of this application is to establish a general approach (rules) for liquid-liquid chromatography separation process development at technical scale (TMB Centrifugal Partition Chromatograph (TMB CPC)), for both batch and continuous separations. We want to develop an approach applicable to a wide range of specific bioproducts separation and purification problems. The method should guide the user in the selection of the mobile and stationary phases (both liquid in liquid-liquid chromatography), operating mode and operating conditions, in order to achieve separation requirements in terms of product purity, recovery and productivity. The input information of the considered approach is: physico-chemical properties of the target component and adjacent impurities, separation purpose (mixture fractionation, target component capturing or polishing) and required separation performances (purity, yield, productivity). All parameters affecting the chromatographic separation efficiency (thermodynamics, hydrodynamics and mass transfer parameters) as well as possible operating modes will be studied systematically through research tasks organized in 5 projects, whose realization is planned in the next 9 years. The aim is to develop a mathematical model based separation process design procedure and minimize the experimental effort for the determination of the parameters governing the process. So, this hardware is crucial for our success.
