We will investigate post-translational arginylation of plant proteins in the model organism Physcomitrella patens, a moss. The aim is to comprehend the role of protein arginylation, its impact on plant development and its influence on the function of selected proteins. We will characterize the changes that occur in response to an arginine-like allelochemical, canavanine. Using modern mass spectrometric tools of proteomics, combined with genetically altered plants lacking the arginylation capability, we will identify proteins that are targets for arginylation as well as factors that are involved in, and may control the modification process. The functional consequences of arginylation of selected proteins will be investigated in vitro.
