This proposal examines the impact of cAMP- and cGMP-dependent modulation of HCN channel activation with regard to the control of network activity and synaptic transmission in thalamic nuclei and hippocampus. In goal 1, a mouse line expressing a cAMP insensitive HCN4 channel will be used to study the role of cAMP regulation of HCN4 for thalamic firing and control of vigilance states. In goal 2, the contribution of HCN2 in GABAergic interneurons in the control of thalamic functions linked to the VB region and the dLGN will be determined. Finally, in goal 3 the role of cGMP-dependent regulation of HCN1 for hippocampal synaptic transmission and spatial memory formation will be investigated.
