This project aims at defining how the HIV-1 pathogenicity factor Nef impairs anterograde transport of Src family kinases to optimize virus spread. The Nef-sensitive pathway acts by a non-classical transport mechanism that requires GTPase-driven solubilization of cargo due to interaction with a lipoprotein chaperone. Building on the identification of candidate molecular targets of Nef with essential roles in this process we will i) define the relevant core transport machinery and determine the molecular lipid and protein composition of involved transport platforms and (ii) dissect the components targeted by Nef and the mechanism of transport disruption.
